Name: ifosfamide
Commercial names: Cyfos, Ifex, Ifosfamidum, Holoxan 1000, Mitoxana, Naxamide
Pharmacological class: alkylating agent
Therapeutic class: antineoplastic
Action: The exact mechanism of ifosfamide has not been determined, but appears to be similar to other alkylating agents. Ifosfamide requires biotransformation in the liver by mixed-function oxidases (cytochrome P450 system) before it becomes active. After metabolic activation, active metabolites of ifosfamide alkylate or bind with many intracellular molecular structures, including nucleic acids. The cytotoxic action is primarily through the alkylation of DNA, done by attaching the N-7 position of guanine to its reactive electrophilic groups. The formation of inter and intra strand cross-links in the DNA results in cell death.

In 2011, ifosfamide is approved to treat:

            ● testicular germ cell tumors that are malignant. It is used in patients who have already been treated with two other types of chemotherapy.

L'action cytotoxique se fait principalement par l'alkylation de l'ADN. La formation de liens dans l’ADN entraîne la mort cellulaire.

En 2011, l'ifosfamide est approuvé pour le traitement:

            ● des tumeurs testiculaires à cellules germinales malignes. Il est utilisé chez les patients qui ont déjà été traités avec deux autres types de chimiothérapie
